采用DNA混池测序法筛选斑点叉尾肌肉生长抑制素(MSTN)基因SNP位点,同时使用SNa Pshot法将筛选到的SNP多态性位点进行分型,最后与生长性状进行关联分析。共筛选到4个SNP位点,即g.3501 T>G、g.3844 T>A、g.4297 C>G、g.4355 G>C。其中,g.3501 T>G位于第一内含子;g.3844 T>A位于第二外显子,并造成密码子GUA变为GAA,氨基酸由缬氨酸(Val)变为谷氨酸(Glu);g.4297 C>G、g.4355 C>G位于第二内含子。关联分析结果表明,SNP位点g.4355 G>C与斑点叉尾的体质量和体长呈显著性负相关,具有GG基因型个体的体质量和体长显著性低于CC型和CG型(P<0.05),该位点可作为斑点叉尾分子育种的候选分子标记。
The SNP loci of myostatin (MSTN) gene were screened by DNA pool hybridization and SNP loci were screened by SNa Pshot method. Finally, the SNPs were compared with the growth traits. A total of four SNPs were screened, that is g.3501 T> G, g.3844 T> A, g.4297 C> G and g.4355 G> C. Among them, g.3501 T> G was located in the first intron; g.3844 T> A was located in the second exon and caused the codon GUA to change to GAA; the amino acid changed from Val to Val (Glu); g.4297 C> G, g.4355 C> G in the second intron. Correlation analysis showed that there was a significant negative correlation between the SNP locus g.4355 G> C and the body weight and body length of Channel catfish, while the body weight and body length of individuals with GG genotype were significantly lower than those of CC and CG (P <0.05), which could be used as a candidate molecular marker for molecular breeding of I.forniochaeta.
