目的 :分析颌面部软组织爆炸伤早期愈合过程中血管内皮生长因子 (VEGF)及其受体 KDR的表达和变化 ,探讨 VEGF在上述损伤愈合过程中的作用。方法 :采用改进家兔颌面部爆炸伤动物模型 ,以DTY- 0 4型雷管为爆炸源 ,复制颌面部爆炸损伤。选用 ABC免疫组化染色法分别检测伤后 6 h、1、2、3、5、7d伤口组织中 VEGF及其受体 KDR的表达 ,并与伤前组比较。结果 :颌面部爆炸伤伤口组织中 VEGF表达在伤后第一周内稳步上升 ,于伤后第一天起和伤前组比较 ,差异明显 ,伤后第 3天开始 ,差异非常显著(P<0 .0 1) ,伤后 7d达峰值。 VEGF受体 KDR的表达则在伤后 3d内与伤前组比较无显著差异 ,伤后 5~ 7d其蛋白质的表达明显加强。结论 :VEGF在颌面部爆炸伤后的表达与创伤愈合过程中新生血管形成的时间相似 ,其受体 KDR的表达也多发生于此时期 ,说明 VEGF参与调节了创伤愈合过程中的血管再生阶段 ,对毛细血管的发生有积极促进作用。
OBJECTIVE: To analyze the expression and changes of vascular endothelial growth factor (VEGF) and its receptor KDR during early healing of maxillofacial soft tissue explosive injury and to explore the role of VEGF in the healing process. Methods: The animal model of maxillofacial explosive injury was modified and the DTY-0 4 detonator was used as the source of explosion to replicate the maxillofacial explosive damage. ABC immunohistochemical staining was used to detect the expression of VEGF and its receptor KDR at 6 h, 1, 2, 3, 5 and 7 days after injury, respectively, compared with the pre-injury group. Results: The expression of VEGF in oral and maxillofacial explosive wounds increased steadily in the first week after injury, and there was a significant difference between the first day after injury and the pre-injury group. On the third day after injury, the difference was significant (P <0. 01), reaching the peak on the 7th day after injury. The expression of VEGF receptor KDR was not significantly different from the pre-injury group within 3 days after injury, and its protein expression was significantly enhanced 5 to 7 days after injury. CONCLUSION: The expression of VEGF in maxillofacial explosive injury is similar to the time of neovascularization during wound healing, and the expression of its receptor KDR also occurs frequently during this period, indicating that VEGF is involved in the regulation of the stage of vascular regeneration during wound healing , The occurrence of capillaries have a positive role in promoting.
