目的:观察单纯性脑震荡(pure cerebral concussion,PCC)致伤后大鼠海马膜结合型前列腺素E2-合酶1(membrane-associated prostaglandin E synthase-1,mPGES-1)在EP2受体抑制剂(AH6809)干预后的表达变化,探讨AH6809对mPGES-1的干预调控作用。方法:采用金属单摆闭合式脑损伤打击装置制备PCC模型,分别于致伤后即刻和12 h腹腔注射AH6809,然后随机分为1、2、3 d和7 d组(n=5),另设相应生理盐水对照组(n=5)。采用免疫组织化学染色和Western Blot方法,观察PCC+AH6809组和生理盐水对照组大鼠海马(CA1~4和上、下齿状回)内m PGES-1的表达情况。结果:免疫组化实验结果显示:在AH6809干预前后均可观察到mPGES-1阳性产物表达于海马CA1~4和上、下齿状回的神经元胞体,呈圆形或卵圆形,胞核不着色,胞浆为棕黄色。与生理盐水对照组相比,AH6809干预后2 d组均出现mPGES-1阳性表达相对减弱,染色渐浅且细胞轮廓不清,3 d组下调最为明显,其中3 d组和7 d组的差异具有显著性(P<0.05)。Western Blot实验结果显示:mPGES-1蛋白出现相似的规律性变化,下降的低峰出现在7 d组,其中2、3、7 d组与对照组相比差异具有显著性(P<0.05)。结论:AH6809对PCC大鼠海马mPGES-1的表达具有一定的干预抑制作用。
OBJECTIVE: To observe the effect of membrane-associated prostaglandin E synthase-1 (mPGES-1) on hippocampal formation induced by pure cerebral concussion (PCC) in EP2 receptor inhibitor (AH6809) after intervention, and to explore the regulatory effect of AH6809 on mPGES-1. Methods: PCC model was prepared by using metal pendulum closed brain injury device. AH6809 was injected intraperitoneally immediately and 12 h after trauma and then randomly divided into 1,2,3 d and 7 d groups (n = 5) Corresponding saline control group (n = 5). Immunohistochemical staining and Western Blot were used to observe the expression of m PGES-1 in hippocampus (CA1 ~ 4 and upper and lower dentate gyrus) in PCC + AH6809 group and saline control group. Results: The results of immunohistochemistry showed that mPGES-1 positive cells were expressed in the hippocampal CA1 ~ 4 and the upper and lower dentate gyrus neurons before and after the intervention of AH6809, showing a round or oval nucleus No coloring, cytoplasm brown. Compared with the saline control group, the expression of mPGES-1 in the 2nd day after the AH6809 intervention was relatively weakened, the staining was gradual and the cell outline was unclear, and the most obvious decrease was seen in the 3-day group, especially in the 3-day and 7-day groups Significant (P <0.05). The result of Western Blot showed that the mPGES-1 protein showed a similar regular pattern, with the decreasing peak appearing in the 7th day. The difference between the 2nd, 3rd, 7th day and the control group was significant (P <0.05). Conclusion: AH6809 can inhibit the expression of mPGES-1 in hippocampus of PCC rats.
