采用碱性蛋白酶酶解酪蛋白制备降血压肽,分别以水解度和ACE抑制活性为指标设计正交试验,确定最佳的酶解条件。最佳条件下的酶解液用超滤的方法进行分离。结果表明,最佳的酶解条件为:底物浓度15%、温度50℃、pH9.5、加酶量0.15%。该条件下的水解度为23.15%,ACE抑制活性为51.82%。经过超滤后,分子量在3 kDa以下的超滤液ACE抑制活性明显提高,起到良好的分离效果。
The antihypertensive peptides were prepared by digestion of casein with alkaline protease. The orthogonal test was designed based on the degree of hydrolysis and ACE inhibitory activity, respectively, and the optimal conditions of enzymatic hydrolysis were determined. Optimal conditions of the enzyme solution by ultrafiltration method for separation. The results showed that the optimal conditions for enzymolysis were as follows: substrate concentration 15%, temperature 50 ℃, pH 9.5, enzyme dosage 0.15%. The degree of hydrolysis under this condition was 23.15%, and the ACE inhibitory activity was 51.82%. After ultrafiltration, the ACE inhibitory activity of ultrafiltrate with molecular weight less than 3 kDa was significantly improved, which has a good separation effect.
