采用微量光度滴定法对花旗松素清除1,1-二苯基-2-苦基苯肼(1,1-Diphenyl-2-picrylhydrazyl radical 2,2-Diphenyl-1-(2,4,6-trinitrophenyl)hydrazyl,DPPH)自由基进行研究,确立微量滴定数学关系,以两者反应的清除计量比R对花旗松素抗氧化能力进行评价,经过数学关系中的公式推导能简便快捷的计算出相应的EC50%值。结果表明,当DPPH初始量分别为1.27×10-7、1.78×10-7 mol时,R值为2.142和2.141,对应的EC50%值为2.967×10-6和4.159×10-6mol/L。在常规实验中,当DPPH初始量均为6.34×10-7 mol时,花旗松素的EC50%值为1.527×10-5mol/L,低于芦丁的EC50%值1.775×10-5 mol/L,说明花旗松素的抗氧化活性比芦丁强。该方法与常规方法结果一致,且该方法具有准确度高、样品用量少、操作简单、成本低等优点,对抗氧化活性物质评价指标的研究具有重要意义。
The photometric titration method was used to determine the effect of 1, 1-diphenyl-2-picrylhydrazyl radical 2,2-diphenyl-1- (2,4,6- trinitrophenyl) hydrazyl, DPPH) free radicals were studied to establish the mathematical relationship between the microtitrelation of the two reaction clearance ratio R on the oxidation of taxifaciens capacity to evaluate, through the mathematical relationship between the deduction formula can be easily and quickly calculate the corresponding EC50% value. The results showed that when the initial volume of DPPH was 1.27 × 10-7 and 1.78 × 10-7 mol respectively, the R values were 2.142 and 2.141, corresponding to the EC50% values of 2.967 × 10-6 and 4.159 × 10-6 mol / L. In routine experiments, when the initial volume of DPPH was 6.34 × 10-7 mol, the EC50% value of Taxifolin was 1.527 × 10-5mol / L and the EC50% value of rutin was 1.775 × 10-5 mol / L, indicating that the antioxidant activity of Taxifolin is stronger than rutin. The method has the same result with the conventional method, and the method has the advantages of high accuracy, less sample amount, simple operation and low cost, and is of great significance to the research on the evaluation index of the antioxidant active material.
