目的:研究表没食子儿茶素没食子酸酯(EGCG)对人雄激素非依赖性前列腺癌细胞端粒酶的抑制作用及可能的作用机制。方法:用不同浓度的EGCG处理体外生长的人雄激素非依赖性前列腺癌PC-3细胞,MTT法检测EGCG对PC-3细胞的抑制作用,TRAP-PCR银染法检测端粒酶活性以及荧光定量PCR法检测人端粒酶逆转录酶mRNA的表达。结果:与对照组相比,EGCG处理组可使存活率明显降低,并随药物剂量的增加呈下降趋势(P<0.01);TRAP-PCR银染法显示,EGCG能明显抑制人雄激素非依赖性前列腺癌PC-3细胞端粒酶的活性,并呈剂量依赖性(P<0.05);荧光定量PCR法显示,被EGCG作用的人雄激素非依赖性前列腺癌PC-3细胞端粒酶逆转录酶mRNA的表达呈下降趋势,并呈剂量依赖性。结论:EGCG可能通过抑制癌细胞的端粒酶的活性来抑制前列腺癌细胞的生长,对癌细胞端粒酶的抑制作用可能是通过抑制端粒酶的关键调节酶-端粒酶逆转录酶来实现的。
Objective: To study the inhibitory effect of epigallocatechin gallate (EGCG) on human androgen-independent prostate cancer telomerase activity and its possible mechanism. Methods: Human androgen-independent prostate cancer PC-3 cells were treated with different concentrations of EGCG. The inhibitory effect of EGCG on PC-3 cells was detected by MTT assay. The telomerase activity and fluorescence Quantitative PCR detection of human telomerase reverse transcriptase mRNA expression. Results: Compared with the control group, the survival rate of EGCG treatment group decreased significantly (P <0.01), and decreased with the dose of EGCG treatment group. TRAP-PCR silver staining showed that EGCG significantly inhibited human androgen-independent (P <0.05). The results of real-time quantitative PCR showed that telomerase reverse was induced by EGCG in human androgen-independent prostate cancer PC-3 cells Enzyme mRNA expression showed a downward trend in a dose-dependent manner. CONCLUSION: EGCG may inhibit the growth of prostate cancer cells by inhibiting the activity of telomerase in cancer cells.
