目的建立1株大鼠源胰腺癌细胞系R-PC。方法利用胰腺包膜下埋二甲基苯并蒽(DMBA)致癌药物的方法建立大鼠胰腺癌模型,用组织块法和胶原酶消化法进行原代培养,观察细胞形态学特征,研究细胞生长状态,测定细胞周期,计数染色体数目,组织化学染色和异种移植。结果经过11个月的体外培养,连续传代80余次;细胞生长迅速,倍增时间为34 h;染色体数目在42~76之间。免疫组织化学染色显示细胞角蛋白、细胞角蛋白19和细胞角蛋白20阳性;异种移植成瘤率为100%。结论R-PC是高度恶性的大鼠胰腺癌细胞系,为体内和体外研究恶性细胞的增值和发展提供了优秀工具。
Objective To establish a rat pancreatic cancer cell line R-PC. Methods The pancreatic cancer model was established by embedding dimethylbenz (a) anthracene (DMBA) into the pancreas under the capsule of the pancreas. The pancreatic cancer model was established by tissue block method and collagenase digestion. The morphological characteristics of the cells were observed. Status, determination of cell cycle, counting of chromosome number, histochemical staining and xenograft. Results After 11 months of in vitro culture, the cells were passaged more than 80 times in succession. The cells grew rapidly and the doubling time was 34 h. The number of chromosomes was between 42 and 76. Immunohistochemical staining showed cytokeratin, cytokeratin 19 and cytokeratin 20 were positive; xenograft tumorigenic rate was 100%. Conclusion R-PC is a highly malignant rat pancreatic cancer cell line and provides an excellent tool for the study of proliferation and development of malignant cells in vitro and in vivo.
