目的 探讨生长抑素类似物奥曲肽(OCT)对胃癌细胞株MKN_(45)生长的体外调节作用。方法 MTT法测定OCT对MKN_(45)细胞增殖的调控;流式细胞术分析OCT对MKN_(45)细胞周期分布的影响;RIA法测定表皮生长因子(EGF)、转化生长因子-α(TGF-α)含量的变化。结果 OCT对MKN_(45)细胞生长有抑制作用,以在1×10~(-6)mol/L浓度时最明显;OCT可诱导MKN_(45)细胞G_0/G_1阻滞,加入OCT后24 h最显著,同期G_2/M期细胞比例亦减少;OCT能抑制胃癌细胞合成EGF、TGF-α的能力。结论 OCT对胃癌生长具有负性调控作用,为胃癌患者应用OCT治疗提供了依据。
Objective To investigate the in vitro regulation of somatostatin analogue octreotide (OCT) on the growth of gastric cancer cell line MKN_ (45). Methods MTT assay was used to determine the effect of OCT on the proliferation of MKN 45 cells. The effect of OCT on the cell cycle distribution of MKN 45 cells was analyzed by flow cytometry (FCM). The expressions of epidermal growth factor (EGF), transforming growth factor- α) content changes. Results OCT could inhibit the growth of MKN_ (45) cells most obviously at the concentration of 1 × 10 -6 mol / L and OCT could induce the G_0 / G_1 arrest of MKN_ (45) cells. After adding OCT for 24 h Most significantly, G 2 / M phase cells decreased over the same period; OCT can inhibit the ability of gastric cancer cells to synthesize EGF, TGF-α. Conclusion OCT negatively regulates the growth of gastric cancer and provides a basis for the treatment of gastric cancer with OCT.
