目的探讨应用广视野钙成像技术观测神经元和胶质细胞的活动。方法采用对流强化法对麻醉雄性大鼠的新皮层进行钙染料染色,应用广视野钙成像技术观测药物诱导的急性癫痫模型中神经元和胶质细胞的活动。结果钙成像的光学脑电图显示癫痫时的神经活动为快速传播的波,其传播范围仅限于致痫灶处,而胶质细胞活动由癫痫起源触发,但是表现为慢速传播的定型波,其传播范围超出神经活动范围。结论广视野钙成像技术更有效地观测神经元和胶质细胞的活动并区别神经元和胶质细胞的网络活动。
Objective To explore the application of wide field calcium imaging technique to observe the activity of neurons and glial cells. Methods The neocortex of anesthetized male rats was stained with calcium dye by convective intensification method. The activity of neurons and glial cells in drug-induced acute epilepsy model was observed by using wide field calcium imaging. Results Calcium-imaging of EEG showed that the neural activity of epilepsy was a fast-propagating wave whose spreading range was limited to the epileptogenic zone. The glial cell activity was triggered by the origin of epilepsy but showed slow-propagating stereotyped waves. Its spread beyond the scope of nerve activity. Conclusion Wide-field calcium imaging technique is more effective in observing the activity of neurons and glial cells and distinguishing the activity of neurons and glial cells.
