目的:研究环腺苷酸 (cyclic adenosine monophosphate, cAMP)、整合素( integrinα 3)在三磷酸腺苷 (adenosin triphosphate, ATP)作用下对人胃癌细胞系( MGC- 803)恶性表型的去恶化作用。方法:应用 ATP作用于人胃癌细胞,①每日计数细胞计算抑制率和绘制生长曲线; ②软琼脂集落形成法检查集落形成能力; ③放免测定法和电镜细胞化学方法检查 cAMP-磷酸二酯酶( cAMP- phosphodiesterase, cAMP- PDEase)活性的改变; ④放免测定法和免疫细胞化学方法检测 cAMP水平的变化;⑤免疫细胞化学方法检测整合素和纽蛋白的表达。结果:① ATP抑制 MGC- 803细胞的增殖和在软琼脂内形成集落的能力,以及 cAMP- PDEase活性; ② ATP提高 MGC- 803细胞内 cAMP水平和增强细胞的整合素和粘着斑-纽蛋白的表达。结论: ATP抑制 MGC- 803细胞增殖并诱导分化; cAMP、整合素和纽蛋白在 ATP对 MGC- 803细胞的恶性表型去恶化过程中起重要的信号调节作用。
Objective: To study the degenerative effects of cyclic adenosine monophosphate (cAMP) and integrin α 3 on the malignant phenotype of human gastric cancer cell line (MGC-803) under the action of adenosin triphosphate (ATP). RESULTS: 1 ATP inhibited the proliferation of MGC-803 cells and the formation of colonies in soft agar, as well as the cAMP-PDEase activity; 2 ATP increased cAMP levels in MGC-803 cells and enhanced the integrins and focal adhesions of neurons. expression. Conclusion: ATP can inhibit the proliferation of MGC-803 cells and induce differentiation. cAMP, integrin and vinculin play an important role in the regulation of malignant phenotype depletion of MGC-803 cells.
