为了研究周期性拉伸应变对人肺上皮细胞DNA合成的影响 ,应用流式细胞术对拉伸应变下的细胞增殖动力学变化进行了分析。结果表明 :在应变为 15 % ,频率为 2 0次 /min、4 0次 /min的拉伸刺激下 ,在 2 4h ,正常人支气管上皮细胞H72 7的增殖活性指数明显降低 ,细胞的DNA合成受到显著抑制 ;4 8h后 ,人肺腺癌细胞A5 49的增殖活性明显降低 ,且拉伸时间越长 ,抑制作用越强。提示周期性拉伸应变抑制人肺上皮细胞的增殖。
In order to investigate the effect of cyclic tensile strain on DNA synthesis in human lung epithelial cells, the kinetics of cell proliferation under tensile strain was analyzed by flow cytometry. The results showed that the proliferation activity index of H72 7 cells in normal human bronchial epithelial cells was significantly decreased at 24 h after the strain was 15%, the frequency was 20 times / min and the tension was 40 times / min. The DNA synthesis After 48 h, the proliferation activity of human lung adenocarcinoma cell A549 was significantly decreased, and the longer the stretching time, the stronger the inhibition effect. Suggesting that cyclic tensile strain inhibits the proliferation of human lung epithelial cells.
