目的观察各种不同细胞外基质成分在体外对7721细胞粘附和迁移的影响。方法运用细胞粘附实验和细胞迁移实验进行观察。结果显示随着纤连蛋白、层连蛋白、Ⅳ型胶原浓度升高,7721细胞在相应的基质上粘附及迁移能力有所上升,低浓度时增强幅度更大。Ⅰ、Ⅲ型胶原在低浓度(<5μg/ml)时表现为轻微诱导粘附和迁移作用,高浓度时(>5μg/ml)抑制粘附和迁移,且随着浓度加大抑制作用更明显。加入各种特异性抗体以及RGD三肽证明上述影响具有基质特异性。结论纤连蛋白、层连蛋白、Ⅳ型胶原可通过诱导肿瘤细胞粘附和迁移,从而促进肿瘤细胞浸润和转移;而Ⅰ、Ⅲ型胶原则相反,可抑制肿瘤细胞的浸润和转移。
Objective To observe the effects of various extracellular matrix components on adhesion and migration of 7721 cells in vitro. Methods Cell adhesion experiments and cell migration experiments were performed. The results showed that as the concentration of fibronectin, laminin, and type IV collagen increased, the adhesion and migration ability of 7721 cells on the corresponding matrix increased, and the increase was greater at low concentrations. Collagen types I and III showed slight adhesion and migration at low concentrations (<5 μg/ml), and inhibited adhesion and migration at high concentrations (>5 μg/ml), and the inhibitory effect was more pronounced with increasing concentrations. . The addition of various specific antibodies and RGD tripeptide demonstrated that the above effects have matrix specificity. Conclusion Fibronectin, laminin and type IV collagen can promote the invasion and metastasis of tumor cells by inducing the adhesion and migration of tumor cells. In contrast, type I and type III colloids can inhibit the infiltration and metastasis of tumor cells.
