目的探讨5-氟尿嘧啶(5-Fu)联合奥沙利铂对SW480细胞成瘤性及其对肿瘤中Galectin-3(半乳糖凝集素-3)表达的影响。方法以人结肠癌SW480细胞建立裸鼠移植瘤模型,实验分5-Fu组,奥沙利铂组,5-Fu+奥沙利铂组和注射等体积生理盐水的对照组。采用免疫组织化学方法检测移植瘤中Galectin-3的表达,以图像信号采集与分析系统进行图像扫描分析。结果 5-Fu组、奥沙利铂组和5-Fu+奥沙利铂组抑瘤率分别为30.53%、34.15%和67.22%;免疫组化显示给药组肿瘤组织中Galectin-3表达明显减弱,Galectin-3在联合用药组肿瘤组织中表达(28.875±7.491)较对照组(281.438±45.639)和单独用药组(157.644±33.721,150.682±25.539)明显减弱,图像灰度值统计分析显示差异均具有统计学意义(P<0.05或P<0.01)。结论 5-Fu和奥沙利铂可抑制肿瘤细胞生长,二者联用抑瘤作用增强。肿瘤的生长体积与Galectin-3的表达具有同向性,提示两者之间可能存在直接或间接的联系。
Objective To investigate the effects of 5-fluorouracil combined with oxaliplatin on tumorigenesis of SW480 cells and its effect on the expression of Galectin-3 (Galectin-3) in tumor cells. Methods Human colon cancer SW480 cells were used to establish xenograft model in nude mice. The experiment was divided into 5-Fu group, oxaliplatin group, 5-Fu + oxaliplatin group and control group with equal volume of saline. The expression of Galectin-3 in the transplanted tumor was detected by immunohistochemical method, and the image scanning analysis was carried out by image signal acquisition and analysis system. Results The inhibitory rates of 5-Fu, oxaliplatin and 5-Fu + oxaliplatin were 30.53%, 34.15% and 67.22%, respectively. Immunohistochemistry showed that the expression of Galectin-3 in the 5-Fu, The expression of Galectin-3 in the tumor tissues of the combination group (28.875 ± 7.491) was significantly weaker than that of the control group (281.438 ± 45.639) and the group administered alone (157.644 ± 33.721 and 150.682 ± 25.539) Statistically significant (P <0.05 or P <0.01). Conclusion 5-Fu and oxaliplatin can inhibit the growth of tumor cells. Tumor growth volume and Galectin-3 expression of the same direction, suggesting that there may be a direct or indirect relationship between the two.
