为了解猪轮状病毒(PoRV)贵州株NSP4基因的序列特征,试验根据Gen Bank上公布的猪轮状病毒NSP4基因序列设计1对特异性引物,扩增一株猪轮状病毒贵州流行株NSP4基因序列并进行序列测定及分析。结果表明:得到的碱基序列(562 bp)与预期结果相符,包含NSP4基因528 bp的开放阅读框,编码175 aa;与已知的14个国内外参考毒株NSP4核苷酸和推导氨基酸序列比较,同源性分别为89.1%~94.1%和95.4%~98.3%;氨基酸系统进化树结果显示,贵州流行株与国内流行株LL36755亲缘关系较近。说明试验成功克隆了PoRV贵州株NSP4基因,获得了其序列,并分析了其序列特征。
In order to understand the sequence characteristics of Porcine Rotavirus (PoRV) Guizhou NSP4 gene, we designed a pair of specific primers based on the sequence of porcine rotavirus NSP4 gene published on Gen Bank to amplify a porcine rotavirus Guizhou strain NSP4 Gene sequences were sequenced and analyzed. The results showed that the obtained nucleotide sequence (562 bp) was consistent with the expected result, including a 528 bp open reading frame of NSP4 gene encoding 175 aa. Compared with the known NSP4 nucleotide and deduced amino acid sequences of 14 domestic and foreign reference strains The homology was 89.1% ~ 94.1% and 95.4% ~ 98.3%, respectively. The results of amino acid phylogenetic tree showed that the Guizhou endemic strains had close genetic relationship with LL36755. This indicated that the NSP4 gene of PoRV Guizhou was successfully cloned and its sequence was obtained. The sequence features of the gene were analyzed.
