目的观察小鼠缺血区脑组织中细胞凋亡情况以及尼莫地平的影响。方法经右侧颈总动脉将尼龙单丝线栓至大脑中动脉造成永久性缺血模型,经尾静脉及腹腔注射尼莫地平,在流式细胞仪上测定凋亡细胞峰。结果细胞凋亡峰于脑缺血后3d出现,单缺组最高,术前给药组最低,术后给药组介于两者之间。单缺组、术前给药组和术后给药组细胞凋亡率分别为(19.24±8.13)%、(6.40±2.15)%和(10.17±3.48)%。结论小鼠大脑中动脉闭塞后细胞凋亡峰于脑缺血后3d出现,尼莫地平可减轻脑缺血区细胞凋亡。
Objective To observe the apoptosis of brain cells in mice and the effect of nimodipine. Methods A permanent ischemic model was established by tethering the nylon monofilament to the middle cerebral artery via the right common carotid artery. The apoptotic cell peak was determined by flow cytometry through tail vein and intraperitoneal injection of nimodipine. Results Apoptosis peak appeared at 3 days after cerebral ischemia, the highest in single defect group was the lowest in preoperative group and the lowest in postoperative group. The apoptotic rates in unipolar group, preoperative group and postoperative group were (19.24 ± 8.13)%, (6.40 ± 2.15)% and (10.17 ± 3.48)%, respectively. Conclusion The peak of apoptosis in the middle cerebral artery after occlusion in mice appears 3d days after cerebral ischemia, and nimodipine can reduce the apoptosis of cerebral ischemia.
