本实验以24只Wistar大鼠的密度梯度离心后的周血淋巴细胞作为观察对象,应用免疫组织化学方法显示EGFR—IR和ACTH—IR,用生物素标记的cRNA探针原位杂交显示EGF的基因表达,此外对比观察了PHA刺激转化的淋巴母细胞和未转化的淋巴细胞的ACTH-IR。结果见到淋巴细胞EGFR-IR呈现阳性、EGF-mRVA信号呈蓝紫色颗粒布满胞质,提示淋巴细胞对EGF可具有自分泌甚至旁分泌效应。ACTH-IR在未转化淋巴细胞呈弱阳性而转化的淋巴母细胞显著增强。
Twenty-four Wistar rats were used to observe the peripheral blood lymphocytes after density gradient centrifugation. EGFR-IR and ACTH-IR were detected by immunohistochemistry. In situ hybridization with biotin-labeled cRNA probe showed that EGF In addition, ACTH-IR of PHA stimulated transformed lymphoblasts and non-transformed lymphocytes was also compared. The results showed that lymphocytes showed positive EGFR-IR, EGF-mRVA signal was blue purple particles covered the cytoplasm, suggesting that lymphocytes can have autocrine EGF or even paracrine effect. ACTH-IR in non-transformed lymphocytes was weakly positive and transformed lymphoblast cells significantly enhanced.
