参照Koh法,改良优化GICC(gastric interstitial Cells of Cajal)的培养方法;采用四唑盐比色实验(MTT法),考察空白血清组、远志血清组、厚朴血清组、远志与厚朴1∶1、1∶2、1∶3配伍血清组对GICC的影响,以优选配比;采用乳酸脱氢酶法(LDH),考察空白血清组、远志血清组、厚朴血清组、远志与厚朴最优配伍血清组对GICC的LDH活力的影响机制。MTT法显示最优配伍组为远志与厚朴1∶2配伍血清组。与空白血清组比较,单味远志血清组(6g/kg)有降低GICC增殖趋势;而与单味远志血清组(6g/kg)比较,远志与厚朴1∶1、1∶2、1∶3配伍血清组,均能极显著促进GICC增殖(P<0.01)。LDH结果显示,与空白血清组比较,单味远志血清组(6g/kg)有降低LDH活力趋势;与单味远志血清组(6g/kg)比较,厚朴含药血清组(6g/kg)、远志与厚朴1∶2配伍血清组能显著增强LDH活力(P<0.05)。综上可见,远志与厚朴配伍能显著促进GICC生长,并提高其存活时间,其机制可能与增强LDH酶活力相关。

Compared with single polygala teniferous group (6g / kg) , Polygalaceae and Magnolia 1: 2 compatibility serum group can significantly enhance LDH activity (P <0.05). In conclusion, compatibility of Polygala and Magnolia officinalis can significantly promote the growth of GICC and increase the survival time of GICC, which may be related to the enhancement of LDH enzyme activity.
