目的探讨凋亡抑制蛋白Bcl-2、凋亡促进蛋白Hrk及细胞周期素E(Cyclin E)在胃癌中的表达及其意义。方法采用免疫组化(S-P)法检测58例胃癌患者的癌组织蜡块标本及正常癌旁组织中Bcl-2蛋白、Hrk蛋白及Cyclin E蛋白的表达情况。结果 Bcl-2蛋白在胃癌组织中阳性表达38例,阳性率为65.5%,癌旁组织中阳性表达14例,阳性率为24.1%,两种组织中Bcl-2蛋白阳性率比较,差异有统计学意义(χ2=20.077,P<0.05)。不同肿瘤分化程度、临床分期及有无淋巴转移胃癌患者中Bcl-2蛋白阳性率比较,差异均有统计学意义(P<0.05);不同年龄、肿瘤直径、Borrmann分型、浸润深度胃癌患者中Bcl-2蛋白阳性率比较,差异均无统计学意义(P>0.05)。Hrk蛋白在胃癌组织中阳性表达12例,阳性率为20.7%,癌旁组织中阳性表达36例,阳性率为62.1%,两种组织中Hrk蛋白阳性率比较,差异有统计学意义(χ2=20.471,P<0.05)。Cyclin E蛋白在胃癌组织中阳性表达34例,阳性率为58.6%,癌旁组织中阳性表达11例,阳性率为18.9%,两种组织中Cyclin E蛋白阳性率比较,差异有统计学意义(χ2=19.206,P<0.05)。Spearman相关分析表明,Bcl-2蛋白与Hrk蛋白呈负相关(r=-0.286,P<0.05),与Cy-clin E蛋白呈正相关(r=0.749,P<0.05)。结论胃癌的形成与多种蛋白的相互作用及基因水平上正负信号转导、调控密切相关,凋亡抑制蛋白Bcl-2、凋亡促进蛋白Hrk及Cyclin E共同参与胃癌的发生发展。
Objective To investigate the expression and significance of apoptosis-inhibiting protein Bcl-2, apoptosis-promoting protein Hrk and cyclin E in gastric cancer. Methods The expressions of Bcl-2, Hrk and Cyclin E proteins in 58 cases of gastric cancer specimens and para-cancerous tissues were detected by immunohistochemistry (S-P). Results The positive rate of Bcl-2 protein in gastric cancer tissues was 38 cases, the positive rate was 65.5%. The positive rate of Bcl-2 protein in paracancerous tissues was 14.1%. There was statistically significant difference between the two groups Significance (χ2 = 20.077, P <0.05). There were significant differences in the positive rate of Bcl-2 protein in different degree of tumor differentiation, clinical stage and lymph node metastasis of gastric cancer (P <0.05). In different age, tumor diameter, Borrmann type, depth of invasion The positive rate of Bcl-2 protein was no significant difference (P> 0.05). The positive rate of Hrk protein in gastric cancer tissues was 12.7% (positive rate was 20.7%). The positive rate of Hrk protein was 36.2% (positive rate was 62.1%). There was significant difference between the two groups (χ2 = 20.471, P <0.05). The positive expression rate of Cyclin E protein in gastric cancer tissues was 34 cases, the positive rate was 58.6%. The positive expression rate of Cyclin E protein in adjacent tissues was 11.9% (positive rate was 18.9%). The positive rate of Cyclin E protein in the two groups was statistically significant χ2 = 19.206, P <0.05). Spearman correlation analysis showed that Bcl-2 protein was negatively correlated with Hrk protein (r = -0.286, P <0.05), and positively correlated with Cy-clin E protein (r = 0.749, P <0.05). Conclusion The formation of gastric cancer is closely related to the interaction of multiple proteins and the positive and negative signal transduction and regulation of gene level. The apoptosis-inhibiting protein Bcl-2, apoptosis-promoting protein Hrk and Cyclin E are involved in the occurrence and development of gastric cancer.
