目的:建立一种测定醋鳖甲中未衍生化甘氨酸(Gly)及脯氨酸(Pro)含量的方法。方法:以反相高效液相色谱-蒸发光散射检测器(HPLC-ELSD)法,采用Prevail C18色谱柱(4.6 mm×250 mm,5μm),以乙腈-0.7%三氟醋酸溶液(含5.0mmol.L-1七氟丁酸)为流动相进行洗脱,洗脱时间为15 min。结果:甘氨酸及脯氨酸峰面积的自然对数与相应浓度的自然对数呈良好的线性关系,线性范围均为为0.1~0.6 g.L-1。平均回收率及RSD分别为Gly 102.5%,RSD 1.9%;Pro 101.2%,RSD 2.5%。结论:该方法不需要专门的氨基酸分析仪和衍生化氨基酸,简便、快速、准确,可作为醋鳖甲药材的质量控制方法。
Objective: To establish a method for the determination of underivatized glycine (Gly) and proline (Pro) content in vinegar. Methods: The compounds were separated on a Prevail C18 column (4.6 mm × 250 mm, 5 μm) using acetonitrile-0.7% trifluoroacetic acid solution (containing 5.0 mmol .L-1 heptafluorobutyric acid) as eluent, the elution time was 15 min. Results: The natural logarithm of glycine and proline peak area showed a good linear relationship with the natural logarithm of corresponding concentration, with the linear range of 0.1-0.6 g · L -1. The average recovery and RSD were Gly 102.5%, RSD 1.9%, Pro 101.2% and RSD 2.5%, respectively. Conclusion: This method does not require a special amino acid analyzer and derivatized amino acids, which is simple, rapid and accurate and can be used as a quality control method for vinegar, turtle and turtle.
