目的观察小鼠内毒素血症IFN-γ、IL-10、IL-2表达特点,探讨IFN-γ、IL-10、IL-2在小鼠内毒素血症发病中的作用及意义。方法选用ICR品系雌性小鼠(8周龄~10周龄),随机分为对照组、模型组,各组30只。每组又随机分为6 h、12 h、24 h 3个时间点,每个时间点有10只动物。内毒素血症模型组(LPS组)腹腔注射LPS溶液9 mg/kg,对照组注入等量的无菌生理盐水,实验组眼球取血,分离血清,用固相夹心法酶联免疫吸附法测定IFN-γ、IL-10、IL-2水平。结果实验组IFN-γ、IL-10、IL-2表达水平较对照组高,差异有统计学意义(P<0.05)。IFN-γ组内比较显示:12 h>6 h>24 h;IL-10组内比较显示:24 h>12 h>6 h;IL-2组内比较显示:6 h>12 h>24 h。结论 IFN-γ、IL-10、IL-2在小鼠内毒素血症的发病机制中可能起着重要作用。
Objective To observe the expression of IFN-γ, IL-10 and IL-2 in endotoxemia mice and to explore the role and significance of IFN-γ, IL-10 and IL-2 in the pathogenesis of endotoxemia in mice. Methods ICR strain female mice (8 weeks to 10 weeks old) were randomly divided into control group, model group and 30 rats in each group. Each group was randomly divided into 6 h, 12 h, 24 h 3 time points, each time point with 10 animals. The LPS group was injected intraperitoneally with LPS solution 9 mg / kg, the control group was injected with the same amount of sterile saline, the eyeballs of the experimental group were taken blood, the serum was separated and determined by solid phase sandwich enzyme-linked immunosorbent assay IFN-γ, IL-10, IL-2 levels. Results The levels of IFN-γ, IL-10 and IL-2 in the experimental group were significantly higher than those in the control group (P <0.05). The intra-IFN-γ group comparison showed that: 12 h> 6 h> 24 h; IL-10 group comparison showed: 24 h> 12 h> 6 h; IL-2 group comparison: 6 h> 12 h> . Conclusion IFN-γ, IL-10 and IL-2 may play important roles in the pathogenesis of endotoxemia in mice.
