目的探讨中枢性甘丙肽(GAL)受体(GALR)对胰岛素敏感性的调节作用。方法18只大鼠随机均分为糖尿病+安慰剂(A)组、糖尿病+GALR拮抗剂M35(B)组和健康对照(C)组。连续侧脑室注射21d后,测量大鼠体重、血糖、胰岛素、葡萄糖输注速率以及脂肪细胞葡萄糖转运蛋白4(GLUT4)mRNA表达。结果与C组比较,A组体重降低、空腹血糖和血清胰岛素水平升高、葡萄糖输注速率降低、GAL mRNA表达水平降低,GLUT4mRNA表达水平降低(P<0.05);与A组比较,B组体重下降,空腹血糖和血清胰岛素水平升高,葡萄糖输注速率降低,GLUT4mRNA表达水平降低(P<0.05)。结论激活中枢GALR能增加脂肪细胞GLUT4表达,促进脂肪细胞对葡萄糖的摄取,增加胰岛素敏感性。
Objective To investigate the regulatory effect of central galanin (GAL) receptor on insulin sensitivity. Methods Eighteen rats were randomly divided into diabetes mellitus + placebo group (A), diabetes + GALR antagonist M35 (B) group and healthy control group (C). After continuous intracerebroventricular injection for 21 days, the body weight, blood glucose, insulin, glucose infusion rate and glucose transporter 4 (GLUT4) mRNA expression were measured. Results Compared with group C, group A had lower body weight, fasting blood glucose and serum insulin levels, lower glucose infusion rate, lower expression of GAL mRNA and lower expression of GLUT4 mRNA (P <0.05). Compared with group A, body weight Decreased, fasting blood glucose and serum insulin levels, glucose infusion rate decreased, GLUT4 mRNA expression decreased (P <0.05). Conclusion Activation of central GALR can increase GLUT4 expression in adipocytes, promote glucose uptake by adipocytes and increase insulin sensitivity.
