As early as 1867, Crambrown et al proposed that changes in the physiological activity of a drug exhibit a corresponding relationship with changes in its chemical structure. Later, around 1900, Overtoae et al. Found that the activity of anesthetic agents with different chemical structures increased with the increase of the lipid-water partition coefficient. The research on the structure-effect relationship after the Second World War began to develop rapidly. In 1961, the analytic relationship between auxin activity and substituent effect was started by the Hansch method.
