目的 测定山东省招远县 1987年 2~ 8月发生的无菌性脑膜炎大流行的病因病毒之W株病毒 (Zhaoyuan W 87)的部分序列 ,分析其与其他肠道病毒的关系。方法 病毒用酚 氯仿抽提RNA ,经逆转录合成cDNA ,经聚合酶链反应 (PCR)扩增 ,产物纯化 ,采用双脱氧链末端终止法在ABI377自动核酸测序仪上测定序列。结果 W株 5′端非编码区、2C区和 3D区的部分序列长度分别为 2 44、2 75和 35 3个核苷酸 ;VP1区全基因为 876个核苷酸 ,编码 2 92个氨基酸 ,与检索到的 2株ECHO2 9核苷酸序列的同源性分别为 82 1%和 81 7% ,氨基酸序列的同源性分别为 95 1%和 94 4% ;与检索到的 1株ECHO2核苷酸序列的同源性为 6 2 3%。种系发生树分析 ,W株在单独的一个分支上 ,与 2株ECHO2 9所在分支较近 ,与ECHO2所在分支较远。结论 目前无法判定W株与ECHO2的关系 ,但可以说W株是ECHO2 9的变异味
Objective To determine the partial sequences of Zhaoyuan W 87, an etiological virus of aseptic meningitis outbreak from January to August 1987 in Zhaoyuan County, Shandong Province, and to analyze its relationship with other enteroviruses. Methods The virus was extracted with phenol and chloroform. The cDNA was synthesized by RT - PCR. The PCR products were purified and sequenced by ABI377 automated nucleic acid sequencer using the dideoxy chain terminator method. Results The partial sequences of the 5 ’untranslated region, the 2C region and the 3D region of W strain were 2, 41, 75 and 35 nucleotides, respectively. The whole gene of VP1 gene was 876 nucleotides encoding a DNA sequence of 2 92 amino acids , Respectively. The homologies of the two nucleotide sequences of ECHO2 9 were 82 1% and 81 7%, respectively. The identities of amino acid sequences were 95 1% and 94 4% respectively. The nucleotide sequence has a homology of 62.3%. Tree phylogenetic tree analysis, W strain on a separate branch, with two branches of ECHO2 9 closer, and ECHO2 where the branch farther. Conclusions At present, it is impossible to determine the relationship between the W strain and ECHO2, but it can be said that the W strain is a mutated taste of ECHO2 9
