目的研究N-去硫酸肝素对大鼠肝缺血再灌注损伤的防治作用。方法 建立肝缺血再灌注损伤大鼠模型,用N-去硫酸肝素对其进行阻断治疗。免疫组化LSAB法检测肝组织P选择素表达。结果 缺血再灌注后大鼠肝细胞显著水肿、空泡变性、炎性细胞浸润,出现大片状坏死灶,血清丙氨酸转移酶和天冬氨酸转移酶水平明显升高(P< 0.01),且P选择素在肝组织中表达。而再灌注前5min静脉注射N-去硫酸肝素,则仅见部分肝细胞轻度水肿、变性及少量的散在点状坏死或局灶性坏死灶伴少量的炎性细胞浸润,未见片状坏死灶,血清酶水平明显减低,肝组织未见P选择素表达。且发现N-去硫酸肝素对凝血指标(PT和KPTT)无明显影响。结论 N-去硫酸肝素对肝缺血再灌注损伤具有防治作用,但对PT和KPTT无明显影响。
Objective To study the preventive and therapeutic effects of N-desulfated heparin on hepatic ischemia-reperfusion injury in rats. Methods The rat model of hepatic ischemia-reperfusion injury was established and blocked with N-desulfated heparin. Immunohistochemistry LSAB method was used to detect P-selectin expression in liver tissue. Results After hepatic ischemia-reperfusion, the hepatocytes were significantly edematous, vacuolar degeneration and infiltration of inflammatory cells with large necrosis of the flakes. Serum levels of alanine aminotransferase and aspartate aminotransferase were significantly increased (P <0.01 ), And P-selectin is expressed in liver tissue. 5 minutes before reperfusion intravenous injection of N- desulfated heparin, only some of the hepatocytes were seen mild edema, degeneration and a small amount of scattered spot necrosis or focal necrosis with a small amount of inflammatory cell infiltration, no necrosis flakes , Serum enzyme levels decreased significantly, no P-selectin expression in liver tissue. N-desulfated heparin was found to have no significant effect on coagulation parameters (PT and KPTT). Conclusion N-desulfated heparin has preventive and therapeutic effects on hepatic ischemia-reperfusion injury but no significant effect on PT and KPTT.
