为探讨神经生长因子(NGF)在附睾中对精子成熟的影响,本研究应用原位杂交和图象分析方法,对成年小鼠附睾中NGF mRNA的分布和相对含量进行检测。实验结果显示:NGF mRNA主要位于附睾管柱状上皮细胞的基底部,原位杂交的信号在附睾管中的分布不均匀,附睾体部杂交信号强于附睾尾部,从附睾管头端至尾端,NGF mRNA含量逐渐下降。实验结果提示精子在进入附睾以后的成熟过程和获得运动能力方面仍然需要NGF的参与。
In order to investigate the effect of nerve growth factor (NGF) on sperm maturation in epididymis, the distribution and relative content of NGF mRNA in epididymis of adult mice were detected by in situ hybridization and image analysis. The results showed that NGF mRNA mainly located in the basal part of the epididymal columnar epithelial cells. The signal of in situ hybridization was unevenly distributed in the epididymis. The hybridization signal of the epididymis was stronger than the tail of the epididymis. From the head to the tail of the epididymis, NGF mRNA levels gradually decreased. The experimental results suggest that sperm still need NGF involvement in the maturation process and gaining exercise capacity after entering the epididymis.
