本实验室用 DL-乙基硫氨酸作为肝脏致癌物来研究食物中胆碱缺乏对大鼠肝癌的发生过程有何影响。在第一组实验中,分别给予大鼠单纯的缺胆碱饲料(GD)或补充胆碱的饮料(GS),第二组实验中分别给予大鼠四种不同饲料(GD、GS 及GD、GS 加0.05%乙基硫氨酸)实验末期处死动物,进行肝切片组织学检查。肝切片和血浆中α-胎儿球蛋白(AFP)免疫萤光镜检,血清中谷丙转氨酸(SGPT)和总胆红质的测定及肝细胞有丝分裂计数。
Our laboratory used DL-ethyl-histidine as a carcinogen to study the effect of choline deficiency in food on the development of hepatocellular carcinoma in rats. In the first experiment, rats were given pure choline aldose (GD) or choline supplementation (GS) respectively. In the second experiment, four different diets (GD, GS and GD, GS plus 0.05% ethionine) were sacrificed at the end of the experiment for histological examination of liver sections. Immunofluorescence microscopy of a-fetoprotein (AFP) in liver sections and serum, determination of glutathione (SGPT) and total bilirubin in serum, and mitotic count of hepatocytes.
