目的:探讨人乳腺癌中真核细胞起始因子-4E(eukaryotic initiation factor4E,eIF-4E)对特异性淋巴管内皮生长因子VEGF-C(vascular endothelial growth factorC)表达和淋巴转移的影响。方法:分别应用Westen-blot法和免疫组化SP法检测57例乳腺癌组织中eIF-4E蛋白水平及VEGF-C表达,VEGFR-3抗体免疫染色计数乳腺癌微淋巴管密度(lymphatic microvessel density,LMVD)。结果:34例淋巴结阳性组织中eIF-4E水平显著高于淋巴结转移阴性组(10·11±0·30,5·94±0·23,t=5·37,P<0·01)。VEGF-C在淋巴结阳性组染色强度明显增高(4·20(0·98),同时VEGFR-3阳性淋巴管密度提高(25·28±5·33),与淋巴结阴性组(2·57±0·88,17·40±4·87)比较差异显著(t=2·18,2·41,P<0·05)。相关分析表明,eIF-4E水平与VEGF-C表达及淋巴结转移呈正相关。结论:eIF-4E可能通过上调VEGF-C的表达诱导乳腺癌淋巴管的生成,进而调控乳腺癌淋巴结转移。eIF-4E可作为判定乳腺癌淋巴结转移的参考指标。
Objective: To investigate the effect of eukaryotic initiation factor 4E (eIF-4E) on the expression of lymphatic endothelial growth factor VEGF-C and lymphatic metastasis in human breast cancer. Methods: The expressions of eIF-4E protein and VEGF-C in 57 cases of breast cancer were detected by Westen-blot and immunohistochemistry respectively. The expression of VEGF-C was detected by immunohistochemical staining of VEGFR-3 and the expression of lymphatic microvessel density LMVD). Results: The levels of eIF-4E in 34 lymph node positive tissues were significantly higher than those in the negative lymph node metastasis group (10.11 ± 0.30, 5.94 ± 0.23, t = 5.37, P <0.01). The staining intensity of VEGF-C in lymph node positive group was significantly higher (4.20, 0.98), and the density of VEGFR-3 positive lymphatic vessel was higher (25.28 ± 5.33) than that in lymph node negative group (2.57 ± 0 · 88,17 · 40 ± 4.87) (t = 2.18,2.41, P <0.05) .The correlation analysis showed that the level of eIF-4E was positively correlated with the expression of VEGF-C and lymph node metastasis .Conclusion: eIF-4E may up-regulate the expression of VEGF-C to induce lymphangiogenesis in breast cancer and further regulate the lymph node metastasis of breast cancer.EIF-4E may be used as a reference index for determining lymph node metastasis in breast cancer.
