目的:观察祛斑汤对雌性C57BL/6J小鼠皮肤α-MSH表达的影响,为探索该方剂治疗黄褐斑机制提供实验依据。方法:将雌性C57BL/6J小鼠随机分为空白对照组、祛斑汤低剂量(10 g/kg)组、中剂量(20 g/kg)组、高剂量(40 g/kg)组、维生素C组。各组小鼠灌胃给药30日后皮肤取材。采用免疫组化法观察皮肤α-MSH表达,并用χ2检验进行统计学分析。结果:祛斑汤高剂量组α-MSH阳性表达率低于空白对照组,差异有统计学意义(P<0.01)。结论:高剂量祛斑汤能降低雌性C57BL/6J小鼠皮肤中α-MSH表达、抑制黑素合成。
Objective: To observe the effect of Quban Decoction on the expression of α-MSH in the skin of female C57BL / 6J mice, and to provide experimental evidence for exploring the mechanism of this recipe in treating melasma. Methods: Female C57BL / 6J mice were randomly divided into blank control group, low dose of Quban Decoction (10 g / kg), medium dose (20 g / kg), high dose (40 g / group. The mice in each group were given gavage for 30 days. Immunohistochemistry was used to observe the expression of α-MSH in skin. Statistical analysis was performed with χ2 test. Results: The positive rate of α-MSH in the high-dose Quban Decoction group was lower than that in the blank control group, with significant difference (P <0.01). Conclusion: High-dose Quban Decoction can reduce the expression of α-MSH in the skin of female C57BL / 6J mice and inhibit melanin synthesis.
