目的:研究水分子通道蛋白1(AQP1)及水分子通道蛋白5(AQP5)在干燥综合征(SS)模型小鼠涎腺的表达及解毒化瘀中药的干预作用。方法:采用诱导法建立SS模型小鼠,并用解毒化瘀中药润燥灵、强的松进行治疗。观察各组小鼠的唾液分泌量变化及小鼠涎腺组织AQP1 mRNA、AQP5 mRNA的表达。结果:与模型组比较,解毒化瘀中药和强的松均能不同程度增加模型小鼠的唾液分泌量,以润燥灵中、高剂量组及强的松组最明显(P<0.05)。模型组AQP1 mRNA含量与空白组比较,差异无显著性意义(P>0.05),经润燥灵及强的松治疗后AQP1 mRNA含量无明显变化(P>0.05)。模型组AQP5 mRNA含量与空白组比较,明显降低(P<0.01),治疗后各组含量均有不同程度升高,以润燥灵中、高剂量组及强的松组最明显(P<0.05)。结论:SS模型小鼠涎腺组织AQP5mRNA表达降低,但对AQP1mRNA无影响,解毒化瘀中药能上调SS模型小鼠涎腺组织AQP5mRNA的表达。
Objective: To study the expression of water channel protein 1 (AQP1) and water channel protein 5 (AQP5) in salivary glands of Sjögren’s syndrome (SS) mouse model and the intervention of detoxification and phlegm drugs. Methods: The SS mice were established by induction method and treated with detoxification and phlegm-removing Chinese herb, Runshaoling and prednisone. The changes of salivary secretion and the expression of AQP1 mRNA and AQP5 mRNA in mouse parotid tissues were observed. RESULTS: Compared with the model group, detoxification, stasis-removing Chinese medicine and prednisone all increased the salivary secretion of model mice in different degrees. The most obvious was in the middle-, high-dose group and prednisone group (P<0.05). The AQP1 mRNA level in the model group was not significantly different from that in the blank group (P>0.05), and the AQP1 mRNA level was not significantly changed after treatment with Runyuling and prednisone (P>0.05). The expression of AQP5 mRNA in the model group was significantly lower than that in the control group (P<0.01). After treatment, the contents of AQP5 in each group increased to different extents, with the most obvious in the Rundrying middle, high dose group and prednisone group (P<0.05). ). Conclusion: The expression of AQP5 mRNA in salivary gland tissue of SS model mice was decreased, but it had no effect on AQP1 mRNA. Detoxification Huayu herbs can upregulate the expression of AQP5 mRNA in salivary gland tissue of SS model mice.
