目的探讨肝X受体(LXR)及其表达基因胆固醇脂转运蛋白(CETP)、环氧化酶-2(COX2)在阻塞性睡眠呼吸暂停低通气综合征(OSAHS)患者中的作用及意义。方法收集2007年8月至2008年8月兰州大学第一医院43例OSAHS患者和45名正常人晨血,经分离培养后得到巨噬细胞,应用逆转录-聚合酶链反应(RT-PCR)凝胶电泳检测法测相关基因表达。结果OSAHS患者LXR、COX2、CETP表达高于对照组(P<0.05);OSAHS患者LXR、COX2、CETP与其呼吸暂停低通气指数(AHI)、最低血氧饱和度(LSaO2)均具有相关性;OSAHS患者LXR、CETP的表达在轻中重3组间存在增强趋势(P<0.05),COX2的表达轻中重3组间没有明显的增强趋势(P>0.05),但在重度与轻度组间差异有统计学意义(P<0.05)。结论OSAHS患者LXR表达显著增高,对CETP、COX2起到调节作用。
Objective To investigate the role of liver X receptor (LXR) and its expression of cholesterol transporter (CETP) and cyclooxygenase-2 (COX2) in patients with obstructive sleep apnea-hypopnea syndrome (OSAHS). METHODS: Forty-three OSAHS patients and 45 healthy volunteers were enrolled in the First Hospital of Lanzhou University from August 2007 to August 2008. The macrophages were isolated and cultured. Reverse transcription-polymerase chain reaction (RT-PCR) Detection of Related Genes by Gel Electrophoresis. Results The expression of LXR, COX2 and CETP in OSAHS patients was higher than that in the control group (P <0.05). The levels of LXR, COX2 and CETP in OSAHS patients were both correlated with AHI and LSaO2. OSAHS The expression of LXR and CETP tended to increase between mild and moderate groups (P <0.05), but there was no obvious increase trend between mild and moderate levels of COX2 expression (P> 0.05) The difference was statistically significant (P <0.05). Conclusion The expression of LXR in patients with OSAHS is significantly increased, which plays a regulatory role on CETP and COX2.
