目的:研究LSA-21型大孔树脂对何首乌二苯乙烯苷的吸附性能及分离纯化工艺参数。方法:采用高效液相色谱法测定2,3,5,4’-四羟基二苯乙烯-2-O-β-D-葡萄糖苷含量,计算LSA-21大孔树脂对二苯乙烯苷的吸附率和解析率。结果:最佳吸附纯化工艺为:最大上样量为1.2 g.g-1(生药量/干树脂),药液浓度为0.2 g.mL-1(相当于生药量),60%乙醇洗脱,洗脱剂用量为6倍树脂量,吸附及洗脱流速均为1.0 mL.min-1(10 g树脂)。结论:LSA-21大孔树脂对何首乌二苯乙烯苷的吸附纯化效果较好,本研究结果可为实际生产提供理论依据。
OBJECTIVE: To study the adsorption properties of LSA-21 macroporous resin to Polygonum multiflorum Thunb. And the purification process parameters. Methods: The content of 2,3,5,4’-tetrahydroxystilbene-2-O-β-D-glucoside was determined by HPLC. The adsorption of stilbene glycoside by LSA-21 macroporous resin Rate and resolution. Results: The optimum adsorption and purification process was as follows: the maximum sample volume was 1.2 gg-1 (raw drug amount / dry resin), the concentration of drug solution was 0.2 g.mL-1 The amount of de-sorbent was 6 times the amount of resin, and the adsorption and elution flow rates were 1.0 mL.min-1 (10 g resin). CONCLUSION: The adsorption and purification of LSH-21 macroporous resin to P. tuberosus is good, and the results of this study can provide theoretical basis for practical production.
