目的观察偏侧帕金森病 (PD)大鼠纹状体多巴胺D2 受体 (D2 R)的功能变化 ,评价12 5I-IBZMD2 R显像的应用价值。 方法应用 6 -羟基多巴胺 (6 -OHDA)建立部分和完全损毁的偏侧PD大鼠模型 ,静脉注射12 5I -IBZM 5 0~ 80 μCi 2h后进行脑组织放射自显影 ;高效液相色谱—电化学法 (HPLC -ECD)检测纹状体多巴胺 (DA)及其代谢产物的含量 ;免疫组化酪氨酸羟化酶 (TH)染色观察黑质及纹状体的细胞和纤维。 结果偏侧PD大鼠损毁侧纹状体12 5I-IBZM摄取增加 ,部分和完全损毁模型的损毁侧较未损毁侧纹状体 /皮层 (ST/CX)比值分别增加了2 3%和 5 4% ;纹状体DA含量分别降低了 39%和 98% ;TH染色可见损毁侧黑质和纹状体TH阳性细胞和纤维明显减少。 结论偏侧PD大鼠模型损毁侧D2 R功能上调 ,且与损毁程度相一致 ;IBZMD2 R显像有助于PD的临床研究
Objective To observe the changes of dopamine D2 receptor (D2 R) in striatum of rats with Parkinson’s disease (PD) and to evaluate the value of 125I-IBZMD2R imaging. Methods Partial and complete destructed hemiparkinsonian rat models were established by 6 - hydroxydopamine (6-OHDA). Brain tissue autoradiography was performed after intravenous 125 I -IBZM 5 0-80 μCi for 2 hours. High performance liquid chromatography The contents of dopamine (DA) and its metabolites in striatum were detected by HPLC-ECD. The cells and fibers of substantia nigra and striatum were observed by immunohistochemical tyrosine hydroxylase (TH) staining. Results Compared with the uninjured side striatum and cortex (ST / CX), the ratio of injured side striatum 125I-IBZM uptake increased and the partial and complete destruction models increased by 23% and 54% respectively %; DA content in the striatum decreased by 39% and 98%, respectively; TH staining showed markedly decreased TH positive cells and fibers in the substantia nigra and striatum. Conclusions The function of D2R on the lesioned side of PD rats is upregulated and consistent with the extent of the lesion; IBZMD2R imaging is helpful for the clinical study of PD
