目的:探讨白藜芦醇诱导K562/ADM细胞凋亡的效应与细胞活性氧水平的关系。方法:应用噻唑蓝(MTT)比色法、光镜、电镜和流式细胞术(FCM)检测细胞凋亡;FCM测定细胞内活性氧(ROS)水平。结果:白藜芦醇显著抑制K562/ADM细胞增殖,形态学呈现典型的凋亡改变;FCM分析显示S期细胞数明显增多,出现S/G2期阻滞;ROS水平升高。结论:白藜芦醇诱导K562/ADM胞凋亡与细胞内活性氧水平升高有关。
Objective: To investigate the effect of resveratrol on the apoptosis of K562 / ADM cells and the relationship between reactive oxygen species (ROS) levels and cell apoptosis. Methods: Cell apoptosis was detected by MTT assay, light microscopy, electron microscopy and flow cytometry (FCM). Intracellular reactive oxygen species (ROS) levels were determined by FCM. Results: Resveratrol significantly inhibited the proliferation of K562 / ADM cells. The morphology of the cells showed typical apoptotic changes. FCM analysis showed that the number of cells in S phase increased significantly, and S / G2 phase arrest and ROS level increased. Conclusion: Resveratrol-induced apoptosis in K562 / ADM cells is associated with an increase in intracellular reactive oxygen species.
