目的研究云南美登木内生真菌Alternaria sp.Ly81的次生代谢产物,并对所得的单体化合物进行抗肿瘤活性筛选。方法采用RP-18,Sephadex LH-20,硅胶等材料进行色谱柱色谱分离化合物,用现代波谱技术鉴定化合物的结构,用MTT法测试单体化合物活性。结果从4 L固体发酵提取物中分离得到9个混合萜类化合物,分别鉴定为tricycloalternarene 2a(1)、tricycloalternarene 1a(2)、tricycloalternarene 1b(3)、tricycloalternarene 2b(4)、tricycloalternarene 3b(5)、tricycloalternarene 4b(6)、tricycloalternarene 5b(7)、tricycloalternarene-H(8)和tricycloalternarene F(9)。结论化合物1~9均为首次从该菌株中分离得到,且首次用Mosher酯反应确定了化合物1的绝对构型,活性筛选结果显示化合物3对Lewis肺癌细胞3LL有强抑制活性。
Aim To study the secondary metabolites of Alternaria sp. Ly81 from the endophytic fungi of Yunnan Mentoring, and to screen the obtained compounds for antitumor activity. Methods Compounds were separated by column chromatography using RP-18, Sephadex LH-20 and silica gel. The structures of compounds were identified by modern spectroscopy. The activity of monomeric compounds was tested by MTT assay. Results Nine mixed terpenoids were isolated from 4 L solid fermentation extracts and identified as tricycloalternarene 2a (1), tricycloalternarene 1a (2), tricycloalternarene 1b (3), tricycloalternarene 2b (4), tricycloalternatene 3b , Tricycloalternarene 4b (6), tricycloalternarene 5b (7), tricycloalternarene-H (8) and tricycloalternarene F (9). Conclusions Compounds 1 to 9 were isolated from this strain for the first time. The absolute configuration of compound 1 was determined by Mosher ester reaction for the first time. Compound 3 showed strong inhibitory activity against Lewis lung carcinoma cell line 3LL.
