利用微波辅助分别以丙酮和含酸无水甲醇为溶剂对柿叶中的单宁进行提取,采用正交试验对提取工艺进行优化。考察了微波功率、反应温度、溶剂浓度、提取时间等因素对单宁提取量的影响。结果表明,以丙酮为提取溶剂时,最佳提取工艺条件为微波功率600 W、反应温度为70℃、溶剂浓度30%、提取时间9 min,由此工艺得单宁的提取量为4.359%;以含酸无水甲醇为提取溶剂时,最佳提取工艺条件为微波功率500 W、反应温度为60℃、溶剂浓度1.4%、提取时间11 min,由此工艺得单宁的提取量为2.957%。以有机溶剂丙酮为提取剂,采用微波辅助提取,单宁提取效果更佳。
Microwave assisted extraction of tannin from persimmon leaves with acetone and acid-free methanol as the solvent respectively, and the orthogonal experiment was used to optimize the extraction process. The effects of microwave power, reaction temperature, solvent concentration, extraction time and other factors on tannin extraction were investigated. The results showed that the optimum extraction conditions were acetone power of 600 W, reaction temperature of 70 ℃, solvent concentration of 30% and extraction time of 9 min. The extraction yield of tannin was 4.359%. When extracting with anhydrous methanol containing methanol, the optimal extraction conditions were microwave power 500 W, reaction temperature 60 ℃, solvent concentration 1.4% and extraction time 11 min. The extraction yield of tannin was 2.957% . With organic solvent acetone as extractant, microwave-assisted extraction, tannin extraction better.
