以一例重症β+地中海贫血患儿的脾DNA组建了λ噬菌体Charon28基因文库。经β-珠蛋白基因组探针筛选出阳性克隆珠,提取其重组体DNA,取其BamHI/1.8kb片段,次级克隆至M13mP18。制备的单链M13mp18重组体DNA经正常与异常的寡核苷酸探针杂交证明该患儿的β地中海贫血基因为β-珠蛋白基因的—28A→G突变体。
The λ-phage Charon28 gene library was constructed from spleen DNA from a patient with severe β + thalassemia. The positive clones were screened by β-globin genomic probe and the recombinant DNA was extracted. The BamHI / 1.8kb fragment was obtained and subcloned into M13mP18. The prepared single-chain M13mp18 recombinant DNA was verified by hybridization of normal and abnormal oligonucleotide probes to show that the β-thalassemia gene of this child was -28A → G mutant of β-globin gene.
