目的研究复合膳食纤维对门静脉高压大鼠肠黏膜屏障功能的保护作用。方法健康雌性Wister大鼠,通过四氯化碳(CCl4)橄榄油复合法建立门静脉高压大鼠模型,建模成功后随机分为A组(单纯模型组)、B组(能全素组)、C组(能全素+膳食纤维组,可溶与不可溶之比为1∶2)、D组(能全素+膳食纤维组,可溶与不可溶之比为2∶1)。分别于建模后1、3、7 d随机取各组3、3、4只大鼠的回肠组织在光镜下观察其病理形态变化,取腹主动脉血检测血浆D-乳酸的表达,取门静脉血检测血浆内毒素的表达。另设正常对照组,同法于第7天取样进行检测。结果建模后各组大鼠的肠黏膜组织结构均有损伤,对照组无损伤;血浆D-乳酸和内毒素表达明显高于对照组,实验组建模后1 d四组组间比较,差异均无统计学意义(血浆D-乳酸:F=0.579,P=0.633;内毒素:F=0.108,P=0.955);3、7 d四组组间比较,差异均高度有统计学意义(血浆D-乳酸3 d:F=39.622,P<0.001,7 d:F=203.834,P<0.001;内毒素3 d:F=95.863,P<0.001,7 d:F=2742.321,P<0.001),且3、7 d各时相B、C、D组大鼠血浆D-乳酸表达和内毒素表达分别与A组比较,差异均有统计学意义(P<0.05),3、7 d C、D组分别与B组比较,差异均有统计学意义(P<0.05),7 d D组与C组比较,差异有统计学意义(P<0.05)。结论复合膳食纤维对门静脉高压大鼠肠黏膜屏障功能有一定的保护作用。
Objective To study the protective effect of compound dietary fiber on intestinal mucosal barrier function in portal hypertensive rats. Methods Healthy female Wister rats were randomly divided into three groups: A group (simple model group), B group (complete plasma group), CCl4 group (CCl4) Group C (vitamins + dietary fiber group, soluble to insoluble ratio of 1: 2), group D (vitamins + dietary fiber group, soluble to insoluble ratio of 2: 1). The ileal tissues of 3, 3, 4 rats in each group were randomly selected at 1, 3 and 7 days after modeling to observe the pathological changes of the ileum. The expression of plasma D-lactic acid was measured by abdominal aorta blood, Detection of Plasma Endotoxin in Portal Vein Blood. Another set of normal control group, the same method in the first 7 days sampling for testing. Results The intestinal mucosa of rats in each group were damaged after injury and the control group was not damaged. The expression of D-lactate and endotoxin in plasma were significantly higher than those in the control group (Plasma D-lactic acid: F = 0.579, P = 0.633; endotoxin: F = 0.108, P = 0.955); the difference between the four groups at 3 and 7 d was highly statistically significant D-lactate 3 d: F = 39.622, P <0.001, 7 d: F = 203.834, P <0.001; Endotoxin 3 d: F = 95.863, P <0.001, 7 d: F = 2742.321, P <0.001) The levels of D-lactate and endotoxin in rats in groups B, C and D on the 3rd and 7th day were significantly different from those in the A group (P <0.05) (P <0.05). Compared with group B, the difference was statistically significant (P <0.05). There was significant difference between group D and group C (P <0.05). Conclusion The compound dietary fiber has a protective effect on intestinal mucosal barrier function in rats with portal hypertension.
