福马林灭活的流感全病毒疫苗(WVV)单独或与霍乱毒素B亚单位(CTB)联合免疫小鼠,能诱导小鼠产生主要组织相容性复合体(MHC)I类细胞毒性T淋巴细胞(CTL),此CTL在人和小鼠体内对限止病毒感染的扩散和清除病毒具有重要作用。 4~8周龄Balb/c小鼠腹腔内接种4.4或44μg流感A/台湾/1/86(HINI)WVV(单独或与5或25μgCTB联合)2次,间隔3周。3周后取小鼠脾细胞体外用A/台湾病毒感染的同系淋巴细胞刺激6~7天。用流感A/台湾和A/上海病毒感染的P815靶细胞评价CTL溶解活性,结果显示:4.4或44μg剂量的WVV诱生的CTL应答等于或大于活流感病毒感染。单用CTB免疫小鼠,未见CTL活性,当用5或25μg CTB与WVV联合免疫时,则能产生比单用WVV免疫更高水平的CTL。 作者还分析了CTL亚型对溶解活性的影响,用单克隆抗体去除Lyt—2~+(CD8~+)T细胞,对P815靶细胞的溶解作用完全消失;而去除L3T4~+(CD4~+)T细胞或自然杀伤细胞,则对细胞溶解作用几乎没有影响。进一步试验显示:足够剂量的流感WVV可诱发小
Fumalin Inactivated Influenza Virus Vaccine (WVV) alone or in combination with cholera toxin B subunit (CTB) can induce mice to produce a major histocompatibility complex (MHC) class I cytotoxic T lymphoid Cells (CTLs), which play an important role in limiting the spread of viral infections and virus clearance in both humans and mice. Four to eight-week-old Balb / c mice were intraperitoneally administered 4.4 or 44 μg of flu A / Taiwan / 1/86 (HINI) WVV (alone or in combination with 5 or 25 μg of CTB) 2 times at 3-week intervals. After 3 weeks, mouse splenocytes were stimulated with A / T virus infected syngeneic lymphocytes for 6 ~ 7 days in vitro. CTL lysis activity was evaluated using P815 target cells infected with influenza A / Taiwan and A / Shanghai virus and the results showed that the WVV-induced CTL response at 4.4 or 44 [mu] g doses was equal to or greater than the live flu virus infection. Mice immunized with CTB alone showed no CTL activity, and CTL was produced when immunized with 5 or 25 [mu] g of CTB in combination with WVV, giving a higher level of immunization than WVV alone.
