用酚/氯仿提取法提取了唇、花和长吻鱼鳍组织的总DNA,利用特异引物进行PCR扩增,得到1140bpCyt b基因片段,纯化后进行测序。所得同源序列用Mega3.1软件分析,并结合鮈亚科其它种类Cyt b基因序列用NJ法和UPGMA法构建系统树。结果表明:(1)三种鱼Cyt b基因片段序列碱基平均差异百分比为13.7%,变异位点201个,转换颠换比为4.6;(2)鲭属三种鱼类成一单系群,唇与花亲缘关系较近。
The total DNA was extracted from the fin tissue of Lip, Scallop, and Anseptha Fabra by phenol / chloroform extraction. The 1140bp Cyt b gene fragment was obtained by PCR using specific primers. After purification, sequencing was carried out. The obtained homologous sequences were analyzed by Mega3.1 software, and the phylogenetic tree was constructed by NJ and UPGMA based on the Cyt b gene sequences of other subfamilies. The results showed that: (1) Cyt b gene fragments of the three species showed an average percentage of base difference of 13.7% with 201 mutation sites and a transversion ratio of 4.6; (2) and flowers kinship closer.
