为探讨体外糖皮质激素对儿童急性特发性血小板减少性紫癜 (AITP)外周血淋巴细胞凋亡的诱导作用 ,对 6例初治的儿童AITP外周血淋巴细胞进行糖皮质激素、糖皮质激素受体拮抗剂 (RU486 )等体外干预培养 ,用DNA原位末端标记技术(Tunel法 )对凋亡细胞进行检测 ,以凋亡指数作为凋亡的定量指标 ,用DNA电泳对细胞凋亡进行定性研究。结果糖皮质激素组的淋巴细胞凋亡指数明显高于正常对照组 ,而在同时加入RU486组 ,其凋亡指数又明显下降 ,与单一糖皮质激素组相比 ,具有显著性差异 ;糖皮质激素组细胞的DNA电泳呈现清晰的“梯状”条带 ,而对照组则未出现。结果表明 ,糖皮质激素具有明显的诱导淋巴细胞凋亡作用 ,而且这一作用可被RU486阻断 ,说明糖皮质激素诱导AITP外周血淋巴细胞凋亡可能是通过糖皮质激素受体途径来实现的 ,从而为治疗提供理论依据。
To investigate the induction of peripheral blood lymphocytes apoptosis in children with acute idiopathic thrombocytopenic purpura (AITP) in vitro, 6 cases of newly diagnosed AITP peripheral blood lymphocytes were treated with glucocorticoid and glucocorticoid Apoptotic cells were detected by Tunel method. Apoptosis index was used as a quantitative indicator of apoptosis. Qualitative study of apoptosis was carried out by DNA electrophoresis. . Results The apoptotic index of glucocorticoid group was significantly higher than that of normal control group, while the apoptotic index of RU486 group was significantly lower than that of normal glucocorticoid group, which was significantly different from that of single glucocorticoid group. Glucocorticoid DNA electrophoresis showed a clear “ladder” band, while the control group did not appear. The results showed that glucocorticoids significantly induced apoptosis of lymphocytes, and this effect can be blocked by RU486, indicating glucocorticoid-induced AITP peripheral blood lymphocyte apoptosis may be achieved through the glucocorticoid receptor pathway , So as to provide a theoretical basis for treatment.
