目的:观察寒燥对慢性阻塞性肺疾病(COPD)模型肺部基质金属蛋白酶-2(MMP-2)m RNA、基质金属蛋白酶-9(MMP-9)m RNA、基质金属蛋白酶抑制剂-1(TIMP-1)m RNA在肺部表达的影响,为寒燥型COPD特点的揭示提供参考。方法:利用气管滴注弹性蛋白酶结合熏烟的方法建立COPD模型,气管滴注弹性蛋白酶、熏烟结合寒燥环境应激的方法建立寒燥型COPD模型,利用Real-time PCR法检测模型肺组织中MMP-2、MMP-9、TIMP-1的m RNA表达。结果:MMP-2 m RNA的表达量COPD寒燥组和COPD模型组均高于对照组(P<0.01),COPD寒燥组高于COPD模型组(P<0.01);MMP-9 m RNA的表达量COPD寒燥组和COPD模型组均高于对照组(P<0.01),COPD寒燥组高于COPD模型组(P<0.05);TIMP-1 m RNA的表达量COPD寒燥组和COPD模型组均高于对照组(P<0.01);MMP-9 m RNA/TIMP-1 m RNA的比值,COPD寒燥组>COPD模型组>对照组。结论:寒燥可以增加COPD模型肺部MMP-2、MMP-9、TIMP-1的m RNA表达,MMP-9 m RNA/TIMP-1 m RNA比值增大,表明气道壁以炎性反应为主,导致组织破坏,形成肺气肿。
OBJECTIVE: To observe the effects of cold and dryness on the expression of matrix metalloproteinase-2 (MMP-2) m RNA, matrix metalloproteinase-9 (MMP-9) m RNA and matrix metalloproteinase inhibitor- 1 in chronic obstructive pulmonary disease (COPD) (TIMP-1) m RNA expression in the lungs, provide a reference for the disclosure of the characteristics of the cold dry COPD. Methods: COPD model was established by intratracheal instillation of elastase combined with fumigation, COPD model was established by tracheal instillation of elastase and fumigation combined with cold and dry environment stress. Real-time PCR was used to detect the expression of lung tissue M-RNA expression of MMP-2, MMP-9 and TIMP-1. Results: The expression of MMP-2 m RNA in COPD cold-drying group and COPD model group were significantly higher than those in control group (P <0.01), COPD cold-drying group was higher than that in COPD model group (P <0.01) The expression of COPD was higher in COPD cold-blooded group and COPD model group (P <0.01), COPD cold-dry group was higher than that in COPD model group (P <0.05) The model group were higher than the control group (P <0.01), the ratio of MMP-9 mRNA / TIMP-1 m RNA, COPD cold group> COPD model group> control group. Conclusion: Cold and dry can increase the m RNA expression of MMP-2, MMP-9 and TIMP-1 in the lungs of COPD model, and increase the ratio of MMP-9 mRNA and TIMP-1 mRNA.
