根据 5- HT受体 13种亚型互补 DNA序列合成相应的特异性引物 ,用 PCR方法对各种 5- HT受体亚型在大鼠三叉神经节及脊髓背根节的表达进行了观察。结果显示 :三叉神经节表达 5- HT1A、5- HT1B、5- HT1D、5- HT2 A、5- HT3、5- HT4 和 5- HT7亚型。背根节表达的 5- HT受体亚型与三叉神经节者相同 ,但这些亚型的电泳条带密度在两种神经节存在着差异。三叉节的 5- HT1D和 5- HT3电泳条带密度最高 ,5- HT1A和 5- HT4 次之 ,5- HT1B、5- HT2 A和 5- HT7较弱 ;背根节的 5- HT3和 5- HT1D电泳条带密度最强 ,5- HT1B和 5- HT2 A次之 ,而 5-HT1A、5- HT4 和 5- HT7则较弱。将各亚型的 PCR阳性产物进一步克隆至 p CR 载体进行序列测定 ,结果表明这些 PCR产物的 DNA序列与已知的各亚型的序列一致。本研究的结果对进一步探讨 5- HT受体在外周感觉信息传递过程中的作用具有一定的意义
According to the complementary DNA sequences of 13 subtypes of 5-HT receptors, specific primers were synthesized and the expression of various 5-HT receptor subtypes in rat trigeminal ganglia and dorsal root ganglion was observed by PCR. The results showed that the trigeminal ganglia express 5- HT1A, 5- HT1B, 5- HT1D, 5- HT2 A, 5- HT3,5- HT4 and 5- HT7 subtypes. The 5-HT receptor subtypes expressed in the dorsal root ganglia are identical to those in the trigeminal ganglion, but the electrophoretic banding density of these subtypes is different between the two ganglia. The 5-HT1D and 5-HT3 electrophoresis band of the trigeminal ganglion had the highest density, followed by 5-HT1A and 5-HT4, and weakened the 5-HT1B, 5-HT2A and 5-HT7. - The strongest HT1D bands were secondary to 5-HT1B and 5-HT2A, while 5-HT1A, 5-HT4 and 5-HT7 were weaker. The PCR positive products of each subtype were further cloned into the p CR vector for sequencing, indicating that the DNA sequences of these PCR products are identical to the known subtypes. The results of this study are of great significance to further explore the role of 5-HT receptors in peripheral sensory information transmission
