药物导致的牙龈过度生长主要发生在三类药物即抗惊厥药、免疫抑制剂及钙离子通道拮抗类降压药剂的应用中。随着这些药物,特别是近年来免疫抑制剂在临床的应用和普及,药物引起牙龈过度生长的现象逐渐受到重视。出于医学伦理的考虑,在人体体内进行药物导致牙龈过度生长的临床实验研究受到诸多制约,因而以实验动物构建病理模型的方法就成为此类研究的重要手段。迄今已在大鼠,比革犬、猕猴、杂种猫等多种动物体内构建了药物导致牙龈过度生长的病理模型,并对其开展了自组织病理学变化到细胞及分子生物学机制等多角度的深入探讨。有鉴于各病理模型在构建方法和研究目的有所不同,研究结论亦不尽一致,本文拟就动物病理模型的构建方法、特点及对药物性牙龈过度生长机制研究的意义进行综述。
Drug-induced gingival overgrowth mainly occurs in the application of three types of drugs, namely anticonvulsants, immunosuppressants and calcium channel blockers. With these drugs, especially in recent years, immunosuppressive agents in the clinical application and popularity, the phenomenon of drug-induced gingival overgrowth gradually attention. Due to medical ethics, there are many limitations in the clinical trials of drug-induced gingival overgrowth in human body. Therefore, the method of constructing pathological model with experimental animals has become an important means of such research. So far, pathological models of drug-induced gingival overgrowth have been established in a variety of animals, such as rats, biogas, macaques, and hybrid cats, and their multi-angles including autophagic pathology and cellular and molecular biological mechanisms have been established In-depth discussion. In view of the different construction methods and research purposes of different pathological models, the research conclusions are not consistent. This paper reviews the construction methods and characteristics of animal pathological models and the significance of the study on the mechanism of drug-induced gingival overgrowth.
