目的 测定卡介菌多糖核酸相对分子质量分布,并观察其热稳定性。方法 采用高效液相分子筛凝胶色谱法测定相对分子质量,并用煮沸或高压强热处理。结果 用该法测定卡介菌多糖核酸相对分子质量分布,有2个色谱峰,第1峰峰值相对分子质量为783 000,第 2峰为 3 200,面积分别为 21%和 79%。热处理后,其第 1峰相对分子质量大约有8%解聚合成第2峰。结论 卡介菌多糖核酸相对分子质量分布集中在3200,约占80%,对热较为稳定。
Objective To determine the relative molecular mass distribution of BCG polysaccharide nucleic acid and observe its thermal stability. Methods The relative molecular mass was determined by high-performance liquid chromatography-gel permeation chromatography (GC-MS), and was treated by boiling or high-pressure heat treatment. Results The relative molecular mass distribution of BCG polysaccharide nucleic acid was determined by this method. There were two peaks, the peak first peak relative molecular mass was 783 000, the second peak was 3 200 and the area was 21% and 79% respectively. After heat treatment, the first peak relative molecular weight of about 8% depolymerization to the second peak. Conclusion The relative molecular weight distribution of BCG-PSN is concentrated in 3200, accounting for about 80%, which is relatively stable to heat.
