目的:探讨I型单纯疱疹病毒(HSV-1)感染对角膜基质金属蛋白酶-2(MMP-2)分布表达的影响。方法:分别收集正常眼球及HSV-1(KOS株)感染后第2,7,14,28d的感染眼球行石蜡包埋,并应用抗MMP-2的多克隆抗体免疫染色角膜切片。结果:感染后第2d,MMP-2的表达比未感染眼增加且表达主要位于浅表基质层及上皮下的炎性细胞中。感染后第7d,MMP-2的表达较第2d减弱。而感染后第14及28d可见角膜基质中及浸润的炎性细胞中尤其是溃疡处可见MMP-2的表达再次显著增加。结论:由角膜细胞及浸润的炎性细胞分泌产生的MMP-2对HSV-1感染所致上皮性角膜炎及角膜溃疡形成可能起重要作用。
Objective: To investigate the effect of HSV-1 infection on the distribution and distribution of corneal matrix metalloproteinase-2 (MMP-2). Methods: The infected eyeballs and HSV-1 (KOS strain) were collected at 2, 7, 14 and 28 days after infection respectively. The corneal slices were immunostained with polyclonal anti-MMP-2 antibody. Results: On the 2nd day after infection, the expression of MMP-2 was higher than that of non-infected eyes and the expression was mainly located in the superficial stromal layer and the inflammatory cells in the epithelium. On the 7th day after infection, the expression of MMP-2 was weaker than that on the 2nd day. On the 14th and 28th day after infection, the expression of MMP-2 in the corneal stroma and infiltrating inflammatory cells, especially ulcers, showed a significant increase again. CONCLUSIONS: MMP-2 secreted by corneal cells and infiltrating inflammatory cells may play an important role in the pathogenesis of keratitis and corneal ulcer caused by HSV-1 infection.
