本文介绍一种快速筛选和准确评价新化合物的药理学特性的麻醉实验动物制备。实验选用体重300~400g雌性豚鼠,二乙巴比妥钠麻醉,恒定正压维持人工呼吸。右颈静脉和颈总动脉插管分别用于给药和记录血压。通过腹中线切口暴露十二指肠,作一小切口插入聚乙烯导管(内径1.0 mm外径1.5mm),导管另端连接注射器,用于十二指肠给药。通过连于放大器的心率计和压力转换器分别记录心率和血压,潮气量变化由连接到中增益前置放大器上的改良Konzett仪测定。暴露膀胱、胆囊、胃、直肠,将聚乙烯导管分别插入膀胱和胆总管;在直肠相距2~3cm处用线扎成一袋,中间作一小切口插入导管;另一导管连接一气球经口通于胃内,各脏器腔内压力分别通过压力转换器记录。
This article presents an anesthetic laboratory animal preparation for rapid screening and accurate evaluation of the pharmacological properties of novel compounds. Experimental selection of body weight 300 ~ 400g female guinea pigs, two barbital sodium anesthesia, constant positive pressure to maintain artificial respiration. Right jugular vein and carotid artery cannulation were used to administer and record blood pressure, respectively. The duodenum was exposed through an incision in the midline and a small incision was made in a polyethylene catheter (1.0 mm id 1.5 mm outer diameter). The other end of the catheter was connected to a syringe for duodenal administration. Heart rate and blood pressure were recorded separately by a heart rate monitor and pressure transducer attached to the amplifier, and the tidal volume changes were measured using a modified Konzett instrument connected to a mid-gain preamplifier. The bladder, gallbladder, stomach and rectum were exposed. The polyethylene catheters were inserted into the bladder and common bile duct, respectively. The rectum was placed 2 to 3 cm from the rectum into a suture. A small incision was inserted in the middle of the duct. Another duct was connected to a balloon via mouth In the stomach, the pressure in each organ is recorded by a pressure transducer.
