用台盼蓝染色及TNS荧光法测定光敏剂DE-03对癌细胞的光动力杀伤作用。结果表明,S_(180),Hepa,U_(14)癌细胞经DE-03 8μg/ml处理和黑光照射20min,杀伤率分别为65.0±2.5%,69.7±2.5%和81.3±2.2%,对S_(180)细胞50%的杀伤浓度为6.93μg/ml。向移植瘤小鼠静脉注射DE-03或YHpD20mg/kg后48h,氨离子泵浦染料激光照射20min,对小鼠S_(180)、U_(14)移植瘤的瘤重抑制率,DE-03为52.2%、47.8%,YHpD为31.3%、40.6%。以耳指数为观察指标,DE-03的光毒反应低于YHpD。
The photodynamic killing effect of photosensitizer DE-03 on cancer cells was determined by trypan blue staining and TNS fluorescence. The results showed that the S_(180), Hepa, U_(14) cancer cells were treated with DE-03 8 μg/ml and irradiated with black light for 20 min. The killing rates were 65.0±2.5%, 69.7±2.5% and 81.3±2.2%, respectively. (180) The 50% killing concentration of cells was 6.93 μg/ml. 48 hours after intravenous injection of DE-03 or YHpD 20mg/kg to tumor-bearing mice, amino-pumped dye laser irradiation for 20 minutes, tumor weight inhibition rate of S 180 and U 14 transplanted tumors in mice, DE-03 was 52.2%, 47.8%, YHpD 31.3%, 40.6%. Using the ear index as an index, the phototoxic response of DE-03 was lower than that of YHpD.
