英国药典(BP)(1980年)用三点校正分光光度法测定维生素A(VA),可修正其吸收带下的线性不相关吸收,但费时较长,如测定“Sevenseas”油胶囊中的VA效价需要3小时才能完成。本文介绍用一阶导数光谱法只需10分钟测定VA的方法。本法为直接记录一阶导数吸收曲线(D_1),读取D_(11)(谷、348nm)和D_(12)(峰、306nm)之间的差ΔD_1。实验证明,VA醋酸酯环己烷溶液在5~25IUml~(-1)浓度范围内,ΔD_1和浓度呈线性关系。此外,在一阶导数曲线D_1(400nm处
The British Pharmacopoeia (BP) (1980) uses a three-point calibration spectrophotometric method to determine vitamin A (VA), which corrects for linear irrelevant absorption under its absorption band, but takes a long time. For example, the determination of VA in “Sevenseas” It takes 3 hours to complete. This article describes the first derivative spectroscopy method that measures VA in 10 minutes. This method directly records the first derivative absorption curve (D_1) and reads the difference ΔD_1 between D_ (11) (valley, 348nm) and D_ (12) (peak, 306nm). Experiments show that, VA acetate cyclohexane solution in the 5 ~ 25IUml ~ (-1) concentration range, ΔD_1 and concentration of a linear relationship.
