目的探讨链球菌抗原(SA)对人外周血单个核细胞来源的树突状细胞(moDCs)成熟的影响。方法体外分离外周血单个核细胞(PBMC),通过重组人粒细胞巨噬细胞集落刺激因子(rhGM-CSF)和重组人白介素4(rhIL-4)诱导moDCs。用链球菌抗原诱导其成熟,ELISA法检测培养上清液中肿瘤坏死因子(TNF-α)和白介素6(IL-6)水平,流式细胞技术检测moDCs表面标志CD80和CD86。结果加入链球菌抗原18h后,moDCs培养上清液的TNF-α和IL-6水平分别为(3 433±363.6)pg/mL和(1 798±49.79)pg/mL,与空白对照比较差异有统计学意义(P<0.000 1)。同时moDCs表面CD80和CD86的表达显著升高。结论链球菌抗原可以诱导人外周血单个核细胞来源的树突状细胞成熟。
Objective To investigate the effect of streptococcal antigen (SA) on the maturation of dendritic cells derived from human peripheral blood mononuclear cells. Methods Peripheral blood mononuclear cells (PBMCs) were isolated in vitro and induced by recombinant human granulocyte macrophage colony-stimulating factor (rhGM-CSF) and recombinant human interleukin-4 (rhIL-4). The maturation was induced by streptococcal antigen. The levels of tumor necrosis factor (TNF-α) and interleukin-6 (IL-6) in culture supernatants were detected by ELISA, and the surface markers of CD80 and CD86 on moDCs were detected by flow cytometry. Results The levels of TNF-α and IL-6 in culture supernatants of moDCs were (3 433 ± 363.6) pg / mL and (1 798 ± 49.79) pg / mL after 18 hours of streptococcal antigen addition, respectively, Statistical significance (P <0.000 1). While the expression of CD80 and CD86 on the surface of moDCs was significantly increased. Conclusion Streptococcal antigens can induce dendritic cells derived from human peripheral blood monocytes to mature.
