目的检测含砷中药雄黄对中国仓鼠肺细胞(CHL细胞)DNA的损伤作用。方法测定雄黄对CHL细胞的半数抑制浓度(IC50),根据IC50设立不同剂量组,进行彗星试验,观察并比较不同组别不同损伤等级细胞数的差异。结果给药干预后,各剂量组及阳性对照组DNA损伤细胞数明显增加,且各剂量组随着给药浓度的增加,Ⅲ、Ⅳ级(中、重度)损伤细胞数增多。结论中药雄黄可引起CHL细胞DNA损伤。
Objective To detect the damage of Chinese traditional medicine containing arsenic to DNA of Chinese hamster lung cells (CHL cells). Methods The median inhibitory concentration (IC50) of realgar against CHL cells was determined. According to the IC50, different dose groups were set up. The comet assay was performed to observe and compare the differences in the number of different grades of cells in different groups. Results After administration, the number of DNA damage cells in each dose group and positive control group increased significantly, and the number of damaged cells in grades III and IV (medium and severe) increased with increasing concentration of each dose group. Conclusion Chinese medicine realgar can cause DNA damage in CHL cells.
